Masteries;

61 - Soul Steal - The only one you should be using, your dots (impale, puncture and jagged strike) will apply a debuff to the mob (Hemophilia) that increases the damage taken from you by 1.5% per stack, stacks up to 3.

62- Revitalize - Everything else in this section is pretty much worthless for assassin, so revitalize is the best choice for the increased healing taken.

63 - You have 2 choices here. You can either go with Juke and Run - Which helps on disconnects with a 12% increased movement speed - Or you can go with Boundless Energy to help with energy drain - Usually if you have Fervor or Living Energy draining isnt an issue, so I went for Juke and Run.

64 - Repetitive Strikes - Increases the Duration of Expose Weakness by 40 seconds and does the most amount of damage compared to the other masteries in this row.

65 - Energy Manipulation - Doesn't really need explaining - Has the highest dps increase. (When parsing use Planar Conservation as this spec can get easily drained)
61 Assassin works really well for full ST fights, its disconnect ability is weak and only has Quick Shot and Head shot thats worth using at range.
Buffs: (You're allowed to have 3 poisons thanks to the 61 point Master of Poisons)

Assassinate (as mentioned) is now your ST spam ability. Quick shot is there incase of disconnects, because of the changes to Assassinate its now no longer worth using Backstab/Puncture, so this will be your main combo point spam. (thanks Bauum)

I don't recommend macro'ing Quick Shot/Headshot, Rift has this lovely bug as to where it can mess up your rotation slightly by not switching back from your ranged weapons to your melee weapons, making it a dps loss to macro if there are disconnects (minimal but noticeable)

Why isnt Serpent Strike in the macro?

Macro'ing Serpent Strike is a really bad idea, you should only be using Serpent Strike when it procs from a critical hit. Which is why if you macro it you risk putting it onto a 20 second cd, rather than an 8 second cd if you use it when it procs.

Impale - DoT finisher (should have a 100% uptime) - ticks once every 2 seconds (will tick at 0 seconds so re-apply as soon as it falls off

Rotation (started at 3 combo points, if you arent at 3 just use your builder macro twice before using impale):
[Stealth] - Pre-cast Exposed Weakness
Start with Jagged Strike - Impale (for damage increase) - Assassinate - CD macro -
During your Poison Malice cooldown block its not worth using Final Blow so just use Assassinate/Serpent Strike/Jagged
Then you will refresh Jagged and Impale as soon as they fall off, using Assassinate and finishers and Serpent Strike, don't use Impale during your Poison Malice CD block
Use your personal cd's as soon as they come off CD - As there will most likely only be one raid CD per boss fight you don't need to try and link up with the supports (unless fights last more than 5 minutes)
Your dots will snapshot the current damaging buff you have active (in this case Call: Razorbeast) so its worth clipping your dots if it means they will carry on past the duration of the buff.
Stealth abilities that stun do not work with Legendary ToD, you need to be in stealth to use things like Debilitating Strike (from assassin) and Ebon Terror (from Nightblade) however, you don't need to use Slip Away as part of your dps rotation anymore, so you can use slip away > Ebon Terror/Debilitating Strike.
Energy Manipulation is boosted by Call: Razorbeast which is why you dont pre-cast it
You have a lot of CC to use in PvP, for example;

Blinding Powder - ST confuse - Lasts 10 seconds (or until damaged) - 20m range - 1m CD

Poison Gas - AoE confuse - Last 6 seconds (or until damaged) - 5m range - 30s CD

Ok so whilst I was testing with Assassin... I found a way to make the rotation literally 4 buttons and still do over 200k dps, you still take the Builder Macro, Cooldown Macro, Serpent Strike and Final blow - Do the same opening rotation from stealth (with jagged) and just mash your builder macro, using CD macro when its off cd, Serpent every time it procs and Final blow at 5 combo points and it performs scary well for how easy it is, not that I advocate this type of rotation im just saying yknow, if you're feeling lazy but still want to do adequate dps in SPE's then... this works xD
